Background Information: | Protein Serine/Threonine Kinase MAPKAPK5 (mitogen-activated protein kinase activated protein kinase 5), also known as p38-regulated/activated protein kinase (PRAK), functions downstream of p38a and p38b in mediating signaling of the p38 pathway. This kinase can be activated in response to cellular stress and pro-inflammatory cytokines through its phosphorylation by MAP kinases including MAPK1/ERK, MAPK14/p38-alpha, and MAPK11/p38-beta. MAPKAPK5 is localized predominantly in the cytoplasm and is constantly shuttled between the cytoplasm and the nucleus. MAPKAPK5 may play a role in balancing other MAPK pathways because over activation of MAPKAPK5 can inhibit Ras mediated cell proliferation and gene activation. MAPKAPK5 inhibition may facilitate cell differentiation, inflammation and cell proliferation.
