PathHunter® Mouse RHO(P23H) ENDO-EA Stable Cell Line Pharmacotrafficking Assay (U2OS)

Background Information:

The PathHunter Mouse RHO(P23H) ENDO-EA Stable Cell Line Pharmacotrafficking Assay (U2OS) utilizes the mouse RHO(P23H) ENDO-EA Pharmacotrafficking cell line. This stable engineered cell line is designed for use in studying pharmacochaperones targeting mouse RHO. This cell line enables assessment of chaperone-based stabilization of mouse RHO(P23H) via detection of rescued membrane-endosome trafficking. This product uses EFC technology. The included cell line overexpresses PK-tagged mutant mouse RHO(P23H), which is misfolded and retained in the ER. The cell line also expresses endosome-localized EA. When a pharmacochaperone stabilizes the mutant mRHO, it is trafficked to the membrane and then internalized to the endosome, producing EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
