Background Information:
The PathHunter Mouse CTLA4 (SHP1) Signaling Stable Cell Line Assay (Jurkat) utilizes the Jurkat mouse CTLA-4 Signaling cell line. The stable engineered cell line is for use in studying drug candidates targeting signaling checkpoints. This cell line enables the assessment of ligand-based activation of mouse CTLA-4 receptors via detection of SH2 domain protein recruitment. This product uses EFC technology. The included cell line overexpresses PK-tagged mouse CTLA-4 target protein and EA-tagged SH2 domain protein. Upon activation of the receptor by its ligand or an agonist antibody, recruitment of a SH2 domain protein to the phosphorylated receptor tail leads to production of the EFC signal. Please note this product expresses the mouse version of CTLA-4.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10 plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
