Background Information:

PathHunter® IL-17 Bioassay Kit (Dimerization) is a quantitative, cell-based assay designed to measure ligand-induced heterodimerization of the IL-17RA and IL-17RC receptor subunits. Activation of the IL-17 signaling pathway by cytokines such as IL-17A and IL-17F plays a central role in chronic inflammatory and autoimmune diseases. Because inhibition of this pathway is a clinically validated therapeutic strategy, reliable functional assays that directly assess target biology are essential for drug discovery, characterization, stability studies and potency testing.
The assay is engineered to replicate the native mechanism of IL-17 receptor activation. Binding of IL-17A or IL-17F to IL-17RC promotes recruitment of IL-17RA, forming the active heterodimeric receptor complex that initiates downstream pro-inflammatory signaling. This bioassay directly quantifies that proximal receptor dimerization event, providing a mechanistically relevant and pathway-specific readout of IL-17 pathway activation. Upon ligand stimulation, engineered cells co-expressing IL-17RA and IL-17RC generate a chemiluminescent signal proportional to receptor interaction. Signal increases in a dose-dependent manner with IL-17A or IL-17F, while neutralizing antibodies, dual IL-17A/F inhibitors, receptor antagonists, and other pathway-targeting therapeutics produce a corresponding reduction in signal. This enables accurate quantification of bioactivity, potency, and functional inhibition.
The kit includes cryopreserved single-use cells, detection reagents, cell plating reagent, assay plates, and agonist control (control-included format), optimized for a 96-well format. A 10-plate kit without control is also available.
