Background Information:
The PathHunter® Human RANK IkB Degradation Cell-Based Assay Kit (U2OS) utilizes the U2OS RANK IκB Degradation Assay Cell Line. The stable engineered cell line is designed for use in studying drug candidates (small molecules or biologics) targeting the RANK NFκB pathway. This cell line enables detection of IκB levels, allowing for assessment of RANK activation-induced IκB degradation. This product uses EFC technology. The cell line included overexpresses untagged RANK and IκB target protein fused to PL. EA is contained in the detection reagent, and complementation of the EA and PL produces EFC signal proportional to the level of IκB in the system. Upon RANK-activation-induced degradation of IκB, EFC signal is lost.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
