Background Information:

The PathHunter® Human IL-18 (IL18R1/IL18RAP) Signaling Reporter Cell-Based Assay Kit (U2OS) utilizes the U2OS IL18R1/IL18RAP Signaling Pathway Reporter cell line. This engineered cell line monitors the activation of specific signaling pathways, utilizing the established PathHunter Enzyme Fragment Complementation (EFC) technology. It can be used for studying drug candidates (small molecules or biologics) affecting the IL-18 pathway. This cell line enables assessment of IL-18 pathway stimulation based on changes in the expression of a NF-kB response element-driven reporter protein. The included cell line overexpresses IL18R1 and IL18RAP, and also expresses PL-tagged reporter protein under the control of a NF-kB-inducible transcriptional response element. Pathway activation induces expression of the PL-tagged protein, allowing detection of the EFC signal upon addition of the EA-containing detection reagent.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
Eurofins DiscoverX optimized Detection Reagents are required for optimal assay performance and must be used with these cell lines in accordance with the cell line license agreed to upon purchase of this cell line assay kit.
