PathHunter® Human AVPR2 (S167T) ENDO-EA Stable Cell Line Pharmacotrafficking Assay (U2OS)

Background Information:

The PathHunter Human AVPR2 (S167T) ENDO-EA Stable Cell Line Pharmacotrafficking Assay (U2OS) utilizes the AVPR2(S167T) ENDO-EA Pharmacotrafficking cell line. The stable engineered cell line is designed for use in studying pharmacochaperones targeting AVPR2. This cell line enables assessment of chaperone-based stabilization of AVPR2(S167T) via detection of rescued membrane-endosome trafficking. This product uses EFC technology. The included cell line overexpresses PK-tagged mutant AVPR2(S167T), which is misfolded and retained in the ER. The cell line also expresses endosome-localized EA. When a pharmacochaperone stabilizes the mutant AVPR2, it is trafficked to the membrane and then internalized to the endosome, producing EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
