Background Information:

The PathHunter® Cyno IL-8 (CXCR1) β-Arrestin Cell-Based Assay Kit (HEK 293) utilizes the Cyno CXCR1 β-Arrestin cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of β-arrestin recruitment following receptor activation for GPCR drug discovery and development programs. This cell line enables the assessment of ligand (e.g., CXCL8/IL8) based activation of cyno CXCR1 GPCR activity via detection of β-arrestin recruitment. This cell line employs Enzyme Fragment Complementation (EFC) technology that combines an overexpressed PK-tagged cyno CXCR1 target receptor and an EA-tagged β-Arrestin 2 to generate a chemiluminescent signal proportional to receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics. All culture and assay reagents, as well as assay detection kits, are available separately to support extended studies of the cell line.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
