Biological Information

Background Information:

MDM2 is an E3 ubiquitin ligase that targets p53 for proteasomal degradation. CK1-delta-dependent phosphorylation modulates MDM2 E3 ligase activity and/or stability, and thereby controls the ubiquitin-dependent turnover of p53. Altered CK1-delta – MDM2 signalling can shift the balance between p53 degradation and stabilization, with major consequences for cell-cycle arrest, DNA-damage responses and apoptosis; processes that are frequently dysregulated in cancer. Targeting CK1-delta offers an indirect route to modulate MDM2 activity, potentially restoring p53 activity in tumours where p53 is wild-type but suppressed by MDM2.
