Background Information:
MDM2 is a RING-type E3 ubiquitin ligase that plays a central role in regulating the tumor suppressor protein p53. Under normal conditions, MDM2 binds directly to the transactivation domain of p53, inhibiting its transcriptional activity and mediating its ubiquitination and proteasomal degradation. This autoregulatory feedback loop ensures that p53 activity remains low in the absence of cellular stress. MDM2 overexpression or amplification is often seen in liposarcomas, leukemias, and some breast and lung cancers.
