Background Information:
The KILR® K562 (Human) Cytotoxicity Assay (cell pool) utilizes the KILR® K562 cell pool. The included cells are a stable, engineered target cell pool, which closely reflects the heterogeneous native protein expression of tumor environments, used to measure direct death mediated by immuno-oncology treatments (e.g., checkpoint modulators, bispecifics, CAR-T, etc.) via Antibody Dependent Cell-mediated Cytotoxicity (ADCC), Complement Dependent Cytotoxicity (CDC), and Antibody Dependent Phagocytosis (ADCP). This product uses EFC technology. The cell pool is a heterogeneous population of K562 cells that overexpresses a ProLabel-tagged housekeeping gene. Once the cells have been lysed by effector cells, ProLabel is released to the media, where EA in the detection kit produces an EFC signal proportional to the level of ProLabel released. This allows for the calculation of the percent lysis of the cell pool.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
