Biological Information

Background Information:

IKKgamma is the regulatory subunit of the inhibitor of IkB kinase (IKK) complex, which activates NF-kappaB resulting in activation of genes involved in inflammation, immunity, cell survival, and other pathways. Binding of IKKgamma to IKKalpha and IKKbeta subunits activates the IKK complex which then phosphorylates the I?Balpha inhibitor protein, leading to its ubiquitin-dependent degradation via the 26S proteasome, and release of NF-kappaB dimers p105 and RELA to translocate to the nucleus and activate transcription of NF-kappaB responsive genes. Due to its central regulatory role in modulating NF-kappaB signalling, IKKgamma has become a focus of drug discovery efforts aimed at cancer and inflammatory diseases. For example, disruption of the IKKgamma-IKKbeta complex by peptides or small molecules has been shown to suppress NF-kappaB activity and impair tumour cell proliferation.
