Background Information:

Idol (Inducible Degrader of the LDL Receptor) is an E3 ubiquitin ligase that regulates cholesterol homeostasis by promoting ubiquitination and lysosomal degradation of the hepatic LDL receptor (LDLR), thereby controlling lipoprotein receptor abundance and circulating LDL cholesterol (LDL-C) levels. Beyond lipid metabolism, Idol also targets related LDLR family members, including VLDLR and ApoER2, implicating it in neuronal development and function. Because LDLR activity is a key determinant of plasma LDL-C, a major risk factor for coronary artery disease, therapeutic strategies that increase hepatic LDLR levels (eg. statins) underpin current cholesterol-lowering treatments. In this regard, modulation of Idol-dependent LDLR degradation represents an attractive alternative or complement to existing therapies designed to enhance LDL clearance and treat hypercholesterolemia.
