Background Information:
The cAMP Hunter™ Human HTR5A (5-HT5A) Gi Cell-Based Assay Kit (CHO-K1) utilizes the Human HTR5A (5-HT5A) cAMP cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of cAMP signaling following receptor activation for drug discovery and development programs. This cell line enables assessment of ligand (e.g. serotonin/5-HT) based activation of HTR5A GPCR activity via detection of decreasing levels of intracellular cAMP. This assay utilizes EFC technology to measure ligand-induced activation by quantifying intracellular cAMP levels. The cells overexpress wild type HTR5A, a naturally Gi/Go coupled GPCR. Upon activation of HTR5A, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. This product leverages a sensitive chemiluminescent cAMP detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
