Background Information:

The PathHunter® U2OS FGR Activity Assay is a stable, engineered cell line for use in studying small molecule drug candidates targeting CTKs. This cell line enables assessment of CTK activation via detection of resulting GHR phosphorylation-induced SH2 domain protein recruitment, mediated by FGR-JAK2 fusion protein. This product uses EFC technology. The included cell line overexpresses PK-tagged relevant cytokine receptor and EA-tagged SH2 domain protein alongside an engineered fusion of the cytokine receptor binding domain of JAK2 (Janus Kinase 2) to the catalytic domain from the CTK of interest, FGR. Upon activation of the cytokine receptor, the CRBD-CTK fusion phosphorylates the receptor, leading to SH2-EA recruitment and production of EFC signal.
This stable engineered cell line is provided as two vials of cryopreserved cells.
