Biological Information
Background Information:
The PathHunter® IGF1R Bioassay Kit (HEK 293) provides an easy-to-use cell-based assay to measure drug potency and detect neutralizing antibodies. This bioassay provides a robust, and highly sensitive functional, cell-based assay that enables the assessment of ligand (e.g. IGF1, IGF2, agonist antibody) based activation of IGF1R receptors via detection of SH2 domain protein recruitment. The included cells overexpress ProLink-tagged IGF1R target protein and EA-tagged SH2 domain protein. Upon activation of the receptor by its ligand or an agonist antibody, recruitment of SH2 domain protein to the phosphorylated receptor tail leads to production of EFC signal. Bioassay kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, control agonist, detection reagent, and assay plates.
The ready-to-use cryopreserved cells have been manufactured to ensure assay reproducibility and faster implementation from characterization to QC lot-release. This bioassay is optimized for a 96-well format and can be further optimized for use in a high-throughput 384-well plate format. The positive control (IGF-1) may not be included with certain configurations of this bioassay kit and may be purchased separately.
