Background Information:
The SPRINTer™ Human BRAF(V600E) Targeted Protein Degradation Cell-Based Assay Kit (A375) utilizes the SPRINTer™ BRAF(V600E) Protein Turnover Biosensor Cell Line (A375) to monitor endogenous BRAF(V600E) protein levels and allow measurement of drug-induced targeted protein degradation (TPD). Genome engineering or editing techniques, such as CRISPR/Cas9, are used to introduce a small tag into the endogenous locus of interest. This allows for the quantitation of changes in BRAF (V600E) levels when cells are treated with therapeutic agents such as targeted protein degraders. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
