PathHunter® Human FLT4 Stable Cell Line Kinase Activity Assay (U2OS)

Background Information:

The PathHunter Human FLT4 Stable Cell Line Kinase Activity Assay (U2OS) utilizes the U2OS FLT4 Activity Assay Cell Line. The stable engineered cell line is intended for use in studying small molecule drug candidates targeting RTKs. This cell line enables the assessment of RTK activation via the detection of resulting FLT4 phosphorylation-induced SH2 domain protein recruitment. The included cell line overexpresses a PK-tagged engineered fusion of the catalytic domain of FLT4 to the extracellular ligand binding domain of EGFR alongside an EA-tagged SH2 domain protein. Upon ligand induced receptor activation, cross-phosphorylation of the receptor leads to SH2-EA recruitment and production of an EFC signal proportional to the degree of receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10 plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
