PathHunter® Human FLT3 Kinase Activity Cell-Based Assay Kit (U2OS)

Background Information:

The PathHunter® Human FLT3 Kinase Activity Cell-Based Assay Kit (U2OS) utilizes the U2OS FLT3 Activity Assay Cell Line. The stable engineered cell line is intended for use in studying small molecule drug candidates targeting RTKs. This cell line enables the assessment of RTK activation via the detection of resulting FLT3 phosphorylation-induced SH2 domain protein recruitment. The included cell line overexpresses a PK-tagged engineered fusion of the catalytic domain of FLT3 to the extracellular ligand binding domain of EGFR alongside an EA-tagged SH2 domain protein. Upon ligand (e.g. induced receptor activation, cross-phosphorylation of the receptor leads to SH2-EA recruitment and production of an EFC signal proportional to the degree of receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
Eurofins DiscoverX optimized Detection Reagents are required for optimal assay performance and must be used with these cell lines in accordance with the cell line license agreed to upon purchase of this cell line assay kit.
