Background Information:
The KILR Human EGFR Stable Cell Line Cytotoxicity Assay (U2OS) utilizes the KILR U2OS-EGFR cell line. The cell line is a stable, engineered target cell line to measure direct cell death mediated by immuno-oncology treatments (e.g., checkpoint inhibitors, checkpoint agonists, bispecifics, CAR-T, and more) via Antibody Dependent Cell-mediated Cytotoxicity (ADCC), Complement Dependent Cytotoxicity (CDC), and Antibody Dependent Cell Phagocytosis (ADCP). This product uses EFC technology. The cell line overexpresses a ProLabel-tagged housekeeping gene as well as untagged EGFR. Once the cells have been lysed by effector cells, ProLabel is released into the media, where EA, contained in the detection kit, produces an EFC signal proportional to the level of PL released. This allows for the calculation of the percent lysis of the cell line.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10 plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
