High Throughput Screening

With 400 validated functional assays, DiscoveRx provides access to the largest menu of available targets for compound library screening. Our services can be used for primary or secondary screening, hit confirmation, or follow-up studies. Choose from our broad portfolio of cell-based assays for GPCRs, Kinases, or NHRs. For GPCR screening, we offer the flexibility of multiple readouts for ß-Arrestin and second messenger detection.

Assay Validation

Prior to screen, all assays are validated according to guidelines set out in the NIH Chemical Genomics Center Assay Guidance Manual. The appropriate controls are included on every plate and Z' values and signal to noise calculations are determined for the entire screen. Any assay that does not meet our specifications or specific criteria set by the customer will be repeated.

Figure 2. Plate uniformity data for a typical PathHunter screen run in antagonist mode. The results show excellent performance for a cell-based assay with an average Z’ of 0.7, 47% inhibition for control antagonist IC₅₀, and overall hit rate of 1.5%.

Our laboratories are fully equipped to process 50,000 wells per day for screens of up to 1,000,000 compounds. Once the initial screen is complete, we can easily perform follow-up work including secondary screening, hit confirmation, and selectivity or rank order profiling.
