PathHunter™ Cell-Based NHR Assays: Protein:Protein Interaction*

The assay detects binding of an agonist or an antagonist to the ligand binding site of NHR of interest and triggering a functional response. Such response is directly measured using interaction of the NHR to Co-activator protein a prerequisite to forming a transcriptional complex. Once bound, complementation occurs between the two β-galactosidase components: the ProLink tag is fused to the C-terminus of the NHR of interest; the Enzyme Acceptor (EA) is attached to SRCP (Steroid response co-activator protein). These components interact only when in close proximity, forming active β-gal enzyme that converts substrate to detectable signal.
