How the cAMP Hunter™ eXpress GPCR Assays Work
GPCR activation following ligand binding initiates a series of second messenger cascades that result in a cellular response. This signaling involves a protein called adenylate cyclase (AC). Gαi and Gαs-coupled receptors modulate cyclic Adenosine Monophosphate (cyclic AMP or cAMP) by either inhibiting or stimulating AC, respectively.
The cAMP Hunter eXpress GPCR assays monitor the functional status of the GPCR by measuring cellular cAMP levels using a homogeneous, gain-of-signal competitive immunoassay based on Enzyme Fragment Complementation (EFC). Thus, the EFC signal generated is directly proportional to the amount of cAMP produced by receptor activation.
