Build Protein Interaction Assays with our ProLink Cloning and Expression Vectors
ProLink Cloning Vectors: ProLink Cloning Vectors are designed such that the gene of interest is cloned in frame with the weakly complementing fragment of β-gal termed ProLink (PK). ProLink is a low affinity tag (~30-fold lower affinity for the Enzyme Acceptor (EA) than the ProLabel) used for construction of protein-protein interaction assays, such as GPCR: β-Arrestin. The ProLink2 (PK2) tag has higher affinity for EA than the original ProLink (PK) tag (~3-fold greater) and is useful for weak protein:protein interactions. For the construction of GPCR cell lines, we have specialized vectors which can improve the S:B ratio. G-protein Receptor Kinase (GRK) phosphorylates GPCRs; facilitating recruitment of arrestins. The Arrestin Recruitment Modifying Sequence (ARMS) is a GRK consensus phosphorylation site deduced from analysis of GPCRs which generate a high signal:background in the PathHunter assay. The addition of ARMS can improve assays which have a low S:B ratio. All ProLink Cloning vectors utilize a minimal CMV promoter for effective expression in mammalian cells.
Choose a ProLabel or ProLink Expression Vector: ProLabel and ProLink Expression Vectors are available with the 4 kDa tag on a wide variety of proteins.
