Forskolin is a labdane diterpene produced by the Indian Coleus plant, Coleus forskohlii. It is commonly used to raise levels of cyclic AMP (cAMP) associated with the study of cell physiology mechanisms, and it activates the enzyme adenyl cyclase to increase intracellular levels of this second messenger, which is necessary for the proper biological response of cells to hormones and other extracellular signals.

Cells were plated in a 384-well plate and incubated overnight at 37°C/5% CO₂ to allow the cells to attach and grow. Cells were then stimulated with a control compound, using the assay conditions described below. Following stimulation, signal was detected using the PathHunter Detection Kit according to the recommended protocol.
