Background Information:

The PathHunter Human MC4R(T162I) ENDO-EA Stable Cell Line Pharmacotrafficking Assay (U2OS) utilizes the MC4R(T162I) ENDO-EA Pharmacotrafficking cell line. The stable engineered cell line is designed for use in studying pharmacochaperones targeting MC4R. This cell line enables assessment of chaperone-based stabilization of MC4R(T162I) via detection of rescued membrane-endosome trafficking. This product uses EFC technology. The included cell line overexpresses PK-tagged mutant MC4R(T162I), which is misfolded and retained in the ER. The cell line also expresses endosome-localized EA. When a pharmacochaperone stabilizes the mutant MC4R, it is trafficked to the membrane and then internalized to the endosome, producing EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
