Biological Information
Background Information:
The PathHunter® Human c-Ret-GFR alpha 2 Functional Kinase Cell-Based Assay Kit (U2OS) utilizes the U2OS c-Ret-GFRα2 Functional Assay Cell Line. This cell based assay is developed to study drug candidates (small molecules or biologics) targeting receptor tyrosine kinases (RTKs). This kit assesses RTK activation by detecting the recruitment of SH2 domain proteins to activated Human c-Ret-GFRα2 dimers. The included cell line overexpresses PK-tagged Human c-Ret, untagged GFRα2, and an EA-tagged SH2 domain protein. Upon ligand (e.g. neurturin)-induced receptor activation, SH2-EA recruitment leads to the production of an EFC signal proportional to the degree of receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
All additional cell culture and assay reagents are available for purchase separately.
