PathHunter® Human CD28 (GRB2) Signaling Stable Cell Line Assay (Jurkat)

Background Information:

The PathHunter Human CD28 (GRB2) Signaling Stable Cell Line Assay (Jurkat) utilizes the CD28 Checkpoint Signaling cell line. The stable engineered cell line is for use in studying drug candidates (small molecules or biologics) targeting signaling checkpoints. This cell line enables the assessment of ligand (e.g., CD80, CD86) based activation of CD28 receptors via detection of SH2 domain protein recruitment. This product uses EFC technology. The included cell line overexpresses PK-tagged CD28 target protein and EA-tagged SH2 domain protein. Upon activation of the receptor by its ligand or an agonist antibody, recruitment of a SH2 domain protein to the phosphorylated receptor tail leads to production of the EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10 plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
