Background Information:

The InCELL Hunter™ Human MEK1 Kinase Target Engagement Cell-Based Assay Kit (A549) utilizes the InCELL Hunter™ A549 MEK1 Kinase Binding cell line. The stable engineered cell line is intended for use in studying small molecule drug candidates that engage with kinases. This cell line enables assessment of compound-engagement-based stabilization of MEK1 via detection of MEK1 levels. This product uses EFC technology. The cell line included overexpresses MEK1's kinase domain fused to PL. In the absence of compound that binds and stabilizes the target, this protein fragment is naturally degraded and no EFC signal is produced. When the compound engages and stabilizes the target, EA contained in the detection reagent produces an EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
Eurofins DiscoverX optimized Detection Reagents are required for optimal assay performance and must be used with these cell lines in accordance with the cell line license agreed to upon purchase of this cell line assay kit.
