Background Information:
The cAMP Hunter™ Human mGlu6 (GRM6) Gi Cell-Based Assay Kit (CHO-K1) utilizes the Human mGlu6 (GRM6) cAMP cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of cAMP signaling following receptor activation for drug discovery and development programs. This cell line enables assessment of ligand (e.g. L-glutamate) based activation of GRM6 GPCR activity via detection of decreasing levels of intracellular cAMP. This assay utilizes EFC technology to measure ligand-induced activation by quantifying intracellular cAMP levels. The cells overexpress wild type GRM6, a naturally Gi/Go coupled GPCR. Upon activation of GRM6, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. This product leverages a sensitive chemiluminescent cAMP detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
