Background Information:
The cAMP Hunter™ Human GIP (GIP Receptor) Gs Cell-Based Assay Kit (CHO-K1) utilizes the Human GIPR cAMP cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of cAMP signaling following receptor activation for drug discovery and development programs. This cell line enables assessment of ligand (e.g. GIP) based activation of GIPR GPCR activity via detection of increasing levels of intracellular cAMP. This assay utilizes EFC technology to measure ligand-induced activation by quantifying intracellular cAMP levels. The cells overexpress wild type GIPR, a naturally Gs coupled GPCR. Upon activation of GIPR, the increase in levels of intracellular cAMP leads to an increased signal from EFC when the cAMP detection kit is applied. This product leverages a sensitive chemiluminescent cAMP detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
Eurofins DiscoverX optimized Detection Reagents are required for optimal assay performance and must be used with these cell lines in accordance with the cell line license agreed to upon purchase of this cell line assay kit.
