Biological Information

Background Information:

XIAP is a multifunctional regulator of programmed cell death and cell-survival pathways. It directly suppresses the catalytic activity of key executioner caspases eg. caspase-3 and caspase-7, by binding their active sites, and maintains caspase-9 in an inactive monomeric state, thereby potently blocking apoptosis at multiple points in the intrinsic and extrinsic death pathways. XIAP also acts as an E3 ubiquitin ligase, ubiquitinating a broad range of signalling proteins including RIPK1, CASP3/7/8/9, survivin (BIRC5) and others, which modulates not only apoptotic signalling but also NF-kappaB, MAPK and Wnt pathways, thereby affecting inflammation and proliferation. Over-expression has been validated in numerous types of cancer cells and has been linked to malignant transformation, increased proliferation and metastasis.
