Background Information:
UbcH4 is a ubiquitin-conjugating enzyme (E2) that plays a central role in the ubiquitin–proteasome system (UPS), which governs targeted protein degradation, signal transduction, and proteostasis. Functionally, UbcH4 receives activated ubiquitin from the E1 ubiquitin-activating enzyme (UBA1) and collaborates with a wide variety of E3 ubiquitin ligases to transfer ubiquitin to specific substrate proteins, forming mono- or polyubiquitin chains that determine substrate fate. Dysregulated UbcH4 activity has been observed in multiple cancers e.g., breast, ovarian, and hepatocellular carcinoma, often correlating with altered ubiquitination of proteins such as p53 and beta-catenin.
