Background Information:

The heterodimeric E2 ubiquitin-conjugating enzyme complex composed of UbcH13 and Uev1A is a specialized E2 that functions with E3 ligases to catalyze K63-linked polyubiquitin chain formation, a non-degradative ubiquitin signal important for intracellular signalling rather than proteasomal degradation. UbcH13–Uev1A acts as an E2 in conjunction with multiple E3 ligases (e.g. TRAF6, TRAF2) to assemble K63-linked ubiquitin chains that act as scaffolds for signal transduction complexes. This K63 ubiquitination is central to activation of the NF-kappa-B pathway, where UbcH13–Uev1A promotes ubiquitination of signalling components such as NEMO. Aberrant dependence on K63 ubiquitination in malignancies such as lymphoma, leukemia, and solid tumours has made this complex an attractive upstream signalling node to modulate oncogenic and inflammatory pathways.
