Background Information:
The PathHunter® Human TrkA Functional Kinase Cell-Based Assay Kit (U2OS) utilizes the U2OS TrkA Functional Assay Cell Line. This cell based assay is developed to study drug candidates (small molecules or biologics) targeting receptor tyrosine kinases (RTKs). This kit assesses RTK activation via the detection of TrkA activation-induced SH2 domain protein recruitment. The included cell line overexpresses PK-tagged Human TrkA and an EA-tagged SH2 domain protein. Upon ligand (e.g. NGF)-induced receptor activation, SH2-EA recruitment leads to the production of an EFC signal proportional to the degree of receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
