Background Information:

The PathHunter® Human Lck Kinase Activity Cell-Based Assay Kit (U2OS) utilizes the U2OS LCK Activity Assay Cell Line. The stable engineered cell line included is intended for use in studying small molecule drug candidates targeting cytosolic kinases. The included cell line overexpresses an engineered fusion of the cytokine receptor binding domain (CRBD) of JAK2 (Janus Kinase 2) to the catalytic domain from the kinase of interest, LCK. It also overexpresses EA-tagged SH2 domain protein alongside a PK-tagged relevant CTK receptor, in this case GHR. Upon activation of the GHR, it is phosphorylated by the CRBD-LCK fusion, leading to SH2-EA recruitment and production of an EFC signal proportional to the degree of receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
