PathHunter® Human Heregulin beta 1 (ErbB2-ErbB3) Stable Cell Line Functional Kinase Assay (U2OS)

Background Information:

The PathHunter Human Heregulin beta 1 (ErbB2-ErbB3) Stable Cell Line Functional Kinase Assay (U2OS) utilizes the U2OS ErbB2-ErbB3 Functional Assay Cell Line. This cell based assay is developed to study drug candidates (small molecules or biologics) targeting receptor tyrosine kinases (RTKs). This kit assesses RTK activation by detecting the recruitment of SH2 domain proteins to activated Human ErbB2-ErbB3 dimers. The included cell line overexpresses PK-tagged Human ErbB2, untagged ErbB3, and an EA-tagged SH2 domain protein. Upon ligand (e.g. heregulin β1)-induced receptor activation, SH2-EA recruitment leads to the production of an EFC signal proportional to the degree of receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
