Background Information:

The PathHunter® Human BCMA Internalization Cell-Based Assay Kit (U2OS) utilizes the BCMA Internalization Cell Line. This stable cell line is engineered to co-express the ProLink™ (PK) tagged BCMA receptor, and an Enzyme Acceptor (EA) tag localized to the endosomes. Ligand (e.g. BAFF, anti-BCMA antibody) binding to the receptor stimulates receptor internalization, resulting in colocalization of the receptor-PK with the EA in the endosomes, and forcing complementation of the two β galactosidase (β-gal) enzyme fragments (EA and PK). The resulting functional β-gal enzyme hydrolyzes substrate to generate a chemiluminescent signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
