Background Information:
The PathHunter® Human Activin A (ACVR1B/BMPR2) Dimerization Cell-Based Assay Kit (U2OS) utilizes the PathHunter® U2OS ACVR1B/BMPR2 Dimerization Cell Line. This assay enables the assessment of ligand (e.g. activin A)-induced dimerization of ACVR1B/BMPR2 pairs. The included cell line employs the PathHunter® Dimerization format using the Enzyme Fragment Complementation (EFC) technology, where the receptor constructs are tagged to enable complementation of the enzyme fragments upon interaction, producing a robust chemiluminescent signal proportional to the dimerization events. The cells overexpress ACVR1B fused to PK and BMPR2 fused to EA, enabling ligand-dependent dimerization measurement upon application of the detection kit.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
