Background Information:

The PathHunter® Human THR beta Nuclear Receptor Protein Interaction Cell-Based Assay Kit (CHO-K1) utilizes the THRβ Protein Interaction cell line. The stable engineered cell line is designed for use in studying drug candidates (small molecules or biologics) affecting NHR activation and recruitment of cofactors. This cell line enables assessment of ligand (e.g., triiodothyronine) based NHR activation via detection of SRCP (steroid receptor co-activator peptide) recruitment. This product uses EFC technology. The included cell line overexpresses PK-tagged THRβ NHR and EA-tagged steroid receptor co-activator peptide (SRCP). Upon activation of THRβ, induced interaction with SRCP leads to production of the EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
