Biological Information

Background Information:

I-kappa-B-alpha is a key cytoplasmic inhibitor of the NF-kappa-B/REL transcription factor complex, which regulates genes involved in inflammation, immune response, cell survival, and oncogenesis. Under basal conditions, I-kappa-B-alpha binds to NF-kappa-B/REL complexes trapping them in the cytoplasm and preventing transcriptional activation. Upon stimulation by pro-inflammatory signals I-kappa-B-alpha becomes phosphorylated and subsequently degraded, releasing RELA to translocate to the nucleus and activate transcription. Stabilizing I-kappa-B-alpha or preventing its degradation may suppress aberrant NF-kappa-B activation, offering therapeutic potential in autoimmune diseases, arthritis, asthma, inflammatory bowel disease and some cancers.
