Background Information:

The cAMP Hunter™ Human OPRK1 (Opioid kappa) Gi Cell-Based Assay Kit (CHO-K1) utilizes the Human OPRK1 (Opioid kappa) cAMP cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of cAMP signaling following receptor activation for drug discovery and development programs. This cell line enables assessment of ligand (e.g. dynorphin A) based activation of OPRK1 GPCR activity via detection of decreasing levels of intracellular cAMP. This assay utilizes EFC technology to measure ligand-induced activation by quantifying intracellular cAMP levels. The cells overexpress wild type OPRK1, a naturally Gi/Go coupled GPCR. Upon activation of OPRK1, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. This product leverages a sensitive chemiluminescent cAMP detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
