Background Information:
The cAMP Hunter™ Human Glucagon (GCG Receptor) Gs Cell-Based Assay Kit (CHO-K1) utilizes the Human GCGR cAMP cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of cAMP signaling following receptor activation for drug discovery and development programs. This cell line enables assessment of ligand (e.g. glucagon) based activation of GCGR GPCR activity via detection of increasing levels of intracellular cAMP. This assay utilizes EFC technology to measure ligand-induced activation by quantifying intracellular cAMP levels. The cells overexpress wild type GCGR, a naturally Gs coupled GPCR. Upon activation of GCGR, the increase in levels of intracellular cAMP leads to an increased signal from EFC when the cAMP detection kit is applied. This product leverages a sensitive chemiluminescent cAMP detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
