NFκB Pathway
Nuclear factor-κB (NF-κB) is a family of highly regulated dimeric transcription factors that play pivotal roles in inflammatory responses and immunological reactions. NFκB pathway can be activated by a variety of stimuli, including TNF-α (tumor necrosis factor α), interleukin 1 (IL-1), T and B cell mitogens, bacterial lipopolysaccharide (LPS) and viral proteins. DiscoveRx offers a simple one-step IκB degradation and NFκB translocation assay to study NFκB pathway. These fully optimized, target-validated cell lines provide you convenience, relevant hits and guaranteed performance for screening, profiling or research applications.
How does the assay work?
DiscoveRx offers a HEK cell line expressing IκB with the ProLabel EFC tag a small peptide epitope (ProLabel) is expressed recombinantly on the intracellular C-terminus of the IκB. In response to TNFα, endogenous TNFa receptor present in HEK cells signal through downstream IκB. Iκb protein is degraded in response to agonist stimulation. The PathHunter® HEK 293 IκB Functional Assay is used with the PL Detection Kit (93-0180) in PathHunter assays for measuring human I-Kappa-B- (IκB, alternate name NFKBIA; NM_020529.2) degradation in response to pathway stimulation.
