Assay Principle: The Cytokine Receptors lack kinase activity and are phosphorylated by Cytosolic Tyrosine Kinases. In this PathHunter® assay approach, a small peptide epitope (ProLink) is expressed recombinantly on the C-terminus of the indicated Cytokine Receptor. One of the many different partner proteins containing SH2 domains is co-expressed with a larger sequence termed enzyme acceptor (EA). This cell line also expresses the Cytosolic Tyrosine Kinase, specified in the product name. Upon ligand-induced activation, the Cytokine Receptor is phosphorylated by the Cytosolic Tyrosine Kinase, causing recruitment of the SH2-containing protein and enabling the two β-gal enzyme fragments to complement and form an active enzyme capable of hydrolyzing a substrate and generating a chemiluminescent signal.
