PathHunter™ Cytosolic Tyrosine Kinases
Unlike receptor tyrosine kinases, the cytokine receptors lack kinase activity and are phosphorylated by cytosolic tyrosine kinases. The interleukin receptors, prolactin, leptin, Erythropoietin (EPO), Thrombopoietin Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF) signal through JAK-STAT pathway, where the receptors are phosphorylated by JAK kinases (which are members of the cytosolic tyrosine kinases). Since, these JAK kinases mediate signalling of several of the receptors involved in autoimmune diseases and cancers, these are viewed as promising new targets for drug discovery. DiscoveRx offers a simple screening friendly cell-based assay for cytokine receptors and the cytosolic tyrosine kinases.
How does the assay work?
To detect the phosphorylation status of cytokine receptors the ProLink is fused to
the c-terminus of the target receptor, and co-expressed with a phosphotyrosine
binding domain fused to the complementing fragment, EA. Upon activation and
phosphorylation of the receptor by the cytosolic kinase, the SH2 domain binds to
the phosphorylated residues. Specificity for a specific kinase can be obtained by
co-expression of specific tyrosine kinases.
